我用FLUO-4 AM检测细胞内钙离子浓度，请问各位战友有什么办法进行钙离子浓度的准确定量啊？

老师，您好！之前看过一个贴，上面是这样说的：1、钙荧光探针负载；取细胞悬液，加Fluo-3-Am至10μmol/L，置二氧化碳培养箱中孵育1h，其间轻微振荡3次。离心，去掉多余的染料。再用无钙缓冲液反复洗涤3次，最后用无钙缓冲液稀释。

2、随机测定每管细胞悬液样品（以下简称样品）的单个细胞的荧光强度，记为F值。

3、加入10%Triton X 100，（破膜用）；加入1mmol/L氯化钙，吹打均匀，孵育1~10min，测定荧光即Fmax值。

4、加入EDTA，20μl（钙螯合剂），孵育1~10min，激发波长488nm测定荧光，即Fmin值。加0.1％triton是增加膜通透性，使细胞外Ca2＋进入细胞内，作为最大值。
然后有一个计算游离钙浓度的公式。并且也有英文文章用了这种方法。那这种方法和您说的这种做标准去曲线的方法有什么区别？哪个更好一点？

不知道公式是怎么样的，既然文章发表了，应该可以参考，至于孰好孰坏，没看到计算原理我也说不好。

Inspectrofluorometer-based assays, it is neces-sary to calibrate at the end of each individualassay by determining the fluorescence intensityof the dye at zero and saturating [Ca2+]i. Fornonratiometric calcium probes such as these,the following equation (Tsien et al., 1982) isused:

                              [Ca2+]i=Kd[(F-Fmin）/（Fmax-F)]

whereKd represents the effective dissociationconstant for calcium-bound fluo-3, Fmax repre-sents the maximum fluorescence (ionophore-treated cells, or detergent-permeabilized cells),Fmin is the fluorescence of the probe in theabsence of calcium, and F is the fluorescenceof the test sample.
就是这样。流式测钙离子浓度9.8章第11页有公式哦。

应该也可以，不过这是将标准曲线直接当成斜向的直线，可能会有偏差。
