倪老师,你好,好久不见。
以前曾经问过您关与检测微粒射门和定量的问题,在FSC-SSC图上圈出0.8μm的标准微球(APC标记)所在的区域,在此区域内分析目的微粒。
文献上说“The diameter of the beads (3 μm, gate B) discriminated
them from the MP population on the FSC-SSC
cytogram. A known number of beads was added to each
sample tube (e.g., 200,000), and the analysis was
stopped when 20,000 beads were counted.”
但是目前有一个问题,我0.8μm的微球是APC标记,两种抗体是PE和APC标记,那3μm的微球是不是要跟抗体的颜色不一样?
不同大小的颗粒,例如3μm和0.8μm如果发光相同,在流式图上能区分开吗?那我能选择PE或者APC标记的3μm的颗粒吗?
PS:因为买不到FITC标记的3μm的微球。只有红色或蓝色的荧光。
