审稿专家提问流式免疫分析里具体细胞数量问题

请教各位老师，审稿专家提问：Furthermore, authors have only presented thepercentage of tumor-infiltrated immune cell analysis in FACS plots, without indicating any specific cell numbers with bar graphs. Asa result, it is challenging to determine whether there were any differences incell infiltration based solely on percentage values.

我看文献里面大家也似乎都是应用百分比形式来展现各组的差异。这位审稿专家似乎是想让我把每组相应的具体细胞数量以柱状图形式展示，请问具体细胞数量哪里可以查到呢？ 没弄明白。不知是否是flowjo里面statistics板块里的count？

这个Reviewer确实有点挑刺的感觉。

要看细胞数量，可直接在flowjo主界面的列表那里，有一列#cells，就是细胞数量

这里面的count是指的颗粒数，严格意义上讲，不是绝对细胞数。如果是要放绝对细胞数量的统计图，可以用体积法或者绝对计数微球，得到这个参数。

用TruCount Beads的管子去测量才能换算出可靠的绝对计数
