In most cases, both B lymphocytes and plasma cells can be demonstrated in WM. Flow cytometry immunophenotyping
analyzes both B and plasmacytic components. The B cells express monotypic surface IgM, light chain and pan-B-cell antigens with moderate-bright intensity, such as CD19, CD20, CD22, and CD79a. The B cells are negative for CD3 and CD103. The plasma
cells express cytoplasmic IgM, light chain, and brightly express plasma cell-associated markers such as CD38 and CD138. The plasma cells are usually positive for CD45 and CD19. (This is in contrast with PCM in which the plasma cells are usually negative for CD19 and weakly positive for CD45.)
