分出的单细胞或者细菌成活率如何？如何提高成活率？

我在分选酵母，然后每次分选出来的酵母成活率都不一致。有时候长得很多，有时候长得很少。我用的是96孔板，是不是可以考虑换更小体积的板？大家做哺乳动物细胞的时候，单细胞成活率如何？体积会影响成活率么？

只要能长，我觉得也不错了。分选后的细胞如果分选时培养基条件好，分选前的细胞状态好，应该是成活率很高的，Purdue上有人说100％：

By PCR, I am able to hit >95% of the wells.  For individual cell lines it can vary.  I have seen as much as >90% with a robust cell type.  Currently with a  Neural Stem Cell line  they are happy with 30%.  The sorter is doing its job if setup correctly.  The problem occurs during culture.  Some cells are damaged and do not “turn” back on for several days.  Some just do not like being a single cell.   Conditioned media seems to help.  I would like as an operator to ensure 100% growth in each well.  I do have to realize that after they leave my sorter it is up to the investigator.  I also like to add a row of more than 1 cell per well.  Sometimes a row of 5 and 10 per well will give good QC results.  If they get no growth from the 10/well then culture conditions need to be addressed.

是什么分析软件里的？是不是直方图的百分比？

哦，是Summit软件里面的，这里的Hist指的是门内细胞占上级门的比例。
