我没用过CANTO II ,但是从下面这个来自http://pathology.jhu.edu/flow/COG%20procedures/Canto_Shutdown_7-20-10.pdf的关机步骤来看,似乎没有涉及到关机液这一说法啊。不知道这一说法从何而来?
从这个关机步骤来看,和CALIBUR的关机步骤基本上差不多,就是先用漂白水冲,再用蒸馏水冲,最后关机。
FACSCanto Shutdown Procedure
1.Double click to open FACS Diva Software.
For Canto A, enter password “pog”.
For Canto II, there is no password.
2.Once in FACS Diva Software, click the “+”(plus) sign next to “DAILY QC” folder.
3.Double click on “Cleaning tube” to open the experiment.
4.For Canto A, make sure the sample probe is in the “UP” position.
For Canto II, there is only one position so no need to adjust.
5.Click the green arrow ONCE next to Tube 1.
6.Place a tube of cleaning solution (i.e. 1:20 Bleach, Contrad, or Coulter Clens) on the Canto and select ‘Acquire Data’. Do Not select ‘Record’! Acquire for 5 minutes.
Note: Bleach tube should be changed at least once a week.
7.For Canto A: Click on ‘Stop Acquiring’ when finished. A message will appear that the computer is “Processing Data”. When “Processing Data” is finished in a few seconds, remove tube by clicking the “Remove Tube” button on the screen and then swing the tube platform arm over and remove tube from the sample probe.
For Canto II: Click on “Stop Acquiring” when finished. The tube platform will automatically come down. Remove tube right away. There is no “Remove Tube” button on Canto II.
8.Repeat steps 5-7 using a deionized water tube for 5 minutes (run water for the same amount of time that you run the cleaning solution).
9.Under ‘Cytometer’ along the menu at the top of page, select ‘Fluidics Shutdown’. A comment will appear:
For Canto A ‘Fluidics shutdown can take up to 7 min’.
For Canto II ‘Fluidics shutdown can take up to 5 min’ .
Click ‘OK’.
10.When Fluidics Shutdown is complete, a message will appear;
“Fluidics Shutdown is complete. It is now safe to turn off the instrument.”
11.Click OK
12.Under ‘File’ at the top of the page, select ‘Quit’. Be sure ‘Exit application only’ is selected, then click ‘OK’.
13.After all functions on the desktop are closed, shut down the computer. Click START at the bottom left corner of the page.
Select ‘Shutdown’ and when the comment “What do you want your computer to do?” appears, be sure the comment in the box is ‘Shutdown”. Click ‘OK’.
14.Turn off the instrument by pressing the green button on the left side of the outside of the FACSCanto.
15.Record the shutdown steps, your initials, and time shutdown in the QC and Maintenance Book.
