slit scan是频率非常高的采集细胞各类参数,而不是一个细胞就扫描一次,所以我觉得担心细胞信号丢失应该是多虑了,应该是更精细才对。
参考下面这段话(来自current protocols in cytometry一书):
If the spot height is made smaller than a celldiameter—i.e., than ~5 µm—cells of differentsizes will have measurably different transittimes through the illuminating beam, yieldingpulses of different widths at the detector(s), andpulse width can then be used to measure cellsize. In the technique of slit-scanning flowcytometry, an extremely small spot height (~2µm) is typically used, and the signal is digitizedat very high rates, allowing a substantialamount of information about cell shape andstructure to be extracted by computer analysis.Until recently, the processing electronics re-quired for slit-scan flow cytometry were toocomplex and expensive to be widely used, butthe hardware and software are now somewhatmore accessible. |
