所以我说他们理解错了，其实你是做了个TO和SSC的二维散点图，他们误认为是一维图，但他们指出的是TO受细胞大小影响大，所以你就用TO和FSC画个图，然后不要圈十字门（因为他们认为随着细胞增大，TO荧光会增强），就按照http://www.flowcyto.cn/bbs/thread-4085-1-1.html中网织血小板最后一个图，画一个斜向的不规则门，圈中阳性部分。

我看了一下您发给我链接，感觉我CD41都是没有必要的。。。。直接用FS 和SS选platelet出来就好了，我又处理了一下，您觉得这样是不是更好啊？

好的，最后我打算这样修改了提交了，也把图片传上来吧，以后万一谁又类似的问题也可以看一下。

对了倪老师，审稿人还提了一个问题，我在文章中写了一句The platelet population in forward (versus side) scattering was confirmed".他要我revised,我没看出问题，请问如何改啊这句

可以试着改成：
The platelet population was gated in FSC(log)/SSC(log) scatter plot.

审稿人还有一个问题。Do you check IPF serially or repeatedly on same day in the same patients, and day by day, if you do this, I'd like to know variability of IPF.
实际上我们并不是所有的患者都重复测量的，只有前面摸索条件的时候部分患者有重复数据，我打算这样回答，倪老师您参谋参谋？
As we have mentioned, the measurement of IPF in our study is by flow cytometry method in a lab of the department of hematology. This lab is a medical laboratory not a research laboratory. Measurement of IPF is a regular item in this lab for assisting in thrombocytopenia diagnosis. This lab achieved the ISO 15189 accreditation. ISO 15189 is a criteria contained particular requirements for quality and competence specifies the quality management system requirements particular to medical laboratories. The result from this lab can be trusted.

Also, we have check the variability of IPF during our study. For the first 10 patients which enrolled in our study, we measured IPF in those patients for 2 times at the same time point. The laboratory coefficient of variation for RP% by flow cytometry is 2.0%.
